How big are the files, if they are 4.36gb x 500 then its going to take a while
4.3gb*500 ~ 2tb? not sure if windows explorer is the best way to copy it, maybe check out xcopy?

In the good old days when moving "large" files I would have said move them in DOS. These days at work we tend to use Linux running on a separate disk to the drives we're moving large files between for speed. Also it helps a lot if they're on separate SATA controllers so they don't share the same system resources, also avoid using one or both drives via USB it will simply bottleneck the transfer speed.

If you must transfer within Windows and if you're using any of the disk services built in to Windows Vista or 7 such as system restore, indexing etc, then transferring large files takes quite a hit in speed. You can speed things up by temporarily disabling these or even by running Windows in Safe Mode, but it will still be slower than the setup we use which gets reasonably close to the theoretical maximum possible for the slower of the two drives or arrays we're moving between.

If you don't have Windows 7 (i.e. XP or Vista) then get a utility called Teracopy, for the fastest possible copying. But there's little difference in transfer speed under win7.

Xcopy is still around? Last time I used Xcopy was on my Amiga 500!

Maybe if you zip'ed up the files (no compression) and did a 1 large file transfer, it works on USB drives.

Two hours sounds about right for 500 mkv files to me and if you don't have two hours spare to transfer them then your never have time to just watch one of them.

Teracopy works well under Windows 7 as well in as much that it frees up the rest of the machine while it is copying. It also allows pause and resume.
If writing to USB, make sure you have Optimise for Speed set rather than for Safe Removal.
