PC wont boot with the card installed but runs fine without the card. I'v tried the card in another pc but only in a normal pci and works fine.

Now to the strange bit I put the card in by mistake without powering off, after the pc rebooted the card was detected under windows. I tried to shutdown and restart but had the same issue no boot so just plugged the card in again while the system was booting and the card shows up again.

Anyone know whats up? mobo is ASUS P5E3 WS Pro card is PCI-X Intel 1000 dual gigabit.

Guessing here. You might have fried something on the motherboard. That was what happened to me when I tested out 'hot plugging'

How far does it get in the boot process? Does it load the bios part of the NIC? The bit where you can press Ctrl+S or something?

It shouldn't matter, but is the slot set for 100MHz or 133MHz? Worse case, use it in a 32bit PCI slot or cover the extra pins. Have you tried a normal 32bit PCI NIC or other device in this PCI-X slot?
